The procedure described allowed a convenient analysis of cyclic nucleotide phosphodiesterase. The different phosphodiesterase forms present in a crude cytosolic preparation from rat heart were separated by isoelectric focusing on a polyacrylamide gel plate. Phosphodiesterase activity bands were rendered evident by a specific staining method. They were then characterized by means of their substrate specificity and their sensitivity to selective phosphodiesterase inhibitors. The correspondence between the stain bands and the previously described activity peaks, obtained by a preparative technique and detected by radioisotopic enzyme assay, was also investigated. 相似文献

Sucrose phosphorylase is an interesting biocatalyst that can glycosylate a variety of small molecules using sucrose as a cheap but efficient donor substrate. The low thermostability of the enzyme, however, limits its industrial applications, as these are preferably performed at 60°C to avoid microbial contamination. Cross-linked enzyme aggregates (CLEAs) of the sucrose phosphorylase from Bifidobacterium adolescentis were found to have a temperature optimum that is 17°C higher than that of the soluble enzyme. Furthermore, the immobilized enzyme displays an exceptional thermostability, retaining all of its activity after 1 week incubation at 60°C. Recycling of the biocatalyst allows its use in at least ten consecutive reactions, which should dramatically increase the commercial potential of its glycosylating activity. 相似文献

Reagent-grade ethylene glycol has been shown to contain substantial amounts of aldehydes, peroxides, iron, and uv-absorbing hydrocarbons. These impurities can be removed by reduction with sodium borohydride, dilution with H2O, passing through a train of four columns, and filtering through a 0.45-micron filter. The product is stable for at least several months and perhaps much longer; storage under nitrogen in acid-washed dark bottles is preferable. Ten liters of 25% (v/v) aqueous ethylene glycol can easily be purified in about 1 week using equipment commonly available in a biochemical laboratory. In a previous study, we demonstrated that Arabidopsis Antioxidant Protein1 (ATX1) plays an essential role in copper (Cu) homeostasis, conferring tolerance to both excess and subclinically deficient Cu. The Cu-binding motif MXCXXC was required for the physiological function of ATX1. In this study, we found that overexpression of ATX1 resulted in hypersensitivity to severe Cu deficiency despite enhancing tolerance to subclinical Cu deficiency. However, overexpression of mutated ATX1, replacing the Cu-binding motif MXCXXC with MXGXXG, abolished the hypersensitivity, for no differences from the wild type under the same conditions. A novel extraction method was developed aiming at increasing the stability of enzymes in organic solvent media. Horseradish peroxidase (HRP), inactivated in a tetrahydrofuran (THF)/water (1:1, v/v), regained and maintained its activity when HRP was extracted by adding a THF/benzene mixture to the original solution. However, the HRP activity was drastically lowered in the enzyme-free blank solution that had been formed by employing the same extraction procedure. Objective: Pleural effusion is common problem, but the rapid and reliable diagnosis for specific pathogenic effusions are lacking. This study aimed to identify the diagnosis based on clinical variables to differentiate pleural tuberculous exudates from other pleural effusions. We also investigated the role of renin-angiotensin system (RAS) and matrix metalloproteinase (MMPs) in the pathogenesis of pleural exudates.Experimental design: The major components in RAS and extracellular matrix metabolism, including angiotensin converting enzyme (ACE), ACE2, MMP-2 and MMP-9 activities, were measured and compared in the patients with transudative (n = 45) and exudative (n = 80) effusions. The exudative effusions were come from the patients with tuberculosis (n = 20), pneumonia (n = 32), and adenocarcinoma (n = 28).Results: Increased ACE and equivalent ACE2 activities, resulting in a significantly increased ACE/ACE2 ratio in exudates, were detected compared to these values in transudates. MMP-9 activity in exudates was significantly higher than that in transudates. The significant correlation between ACE and ACE2 activity that was found in transudates was not found in exudates. Advanced analyses showed significantly increased ACE and MMP-9 activities, and decreased ACE2 activity in tuberculous pleural effusions compared with those in pneumonia and adenocarcinoma effusions. The results indicate that increased ACE and MMP-9 activities found in the exudates were mainly contributed from a higher level of both enzyme activities in the tuberculous pleural effusions.Conclusion: Interplay between ACE and ACE2, essential functions in the RAS, and abnormal regulation of MMP-9 probably play a pivotal role in the development of exudative effusions. Moreover, the ACE/ACE2 ratio combined with MMP-9 activity in pleural fluid may be potential biomarkers for diagnosing tuberculous pleurisy. 相似文献

The interrelation of Alpine topography with the micro − and mesobiota is still poorly understood. We investigated the effects of ground cover type and slope exposure on the soil microbial biomass (double-stranded DNA, dsDNA) and abundances (real time PCR, qPCR); hydrolytic enzyme activities; and enchytraeid community structure in top soils (2.5-cm increments depth) in subalpine forests in the Italian Alps. Dominant ground covers were grass, moss, litter and woody debris at the north- and the south-facing slopes. The autochthonous soil microbiota (bacteria, fungi and archaea) was quantified by qPCR in the extracellular (eDNA) and intracellular fraction (iDNA) of the total soil DNA pool. A higher eDNA/iDNA ratio indicative of lower microbial activity was recorded in the deepest layer of the grass plots at the north-facing slope. This can be related to a lower degradation of eDNA and/or to an accumulation of eDNA with increasing depth as a result of leaching. The exposure effect was enzyme-specific and higher activities occurred under woody debris primarily at the south-facing slope. These plots also showed a higher nutrient content and a greater microbial biomass assessed as dsDNA yields. Total microannelid abundance was elevated on north-facing slopes on account of strong acidity indicator species. This was related to soil pH being one unit lower compared to the south-facing slope. The thickness of the organic layer (OL + OF + OH) was elevated at the north-facing slope due to a considerably thicker OH-horizon. The vast majority of microannelids at this slope occurred in the organic layer, while at south exposure they were almost evenly distributed between the organic layer and the mineral soil (A-horizon). The radical-scavenging reaction of fisetin, a natural antioxidant found in strawberries, is known to proceed via hydrogen transfer to produce a fisetin radical intermediate. Thus, introduction of an electron-donating group into the fisetin molecule is expected to stabilize the radical, leading to enhanced radical-scavenging activity. In this study, fisetin derivatives in which methyl substituents were introduced at the ortho positions relative to the catechol hydroxyl groups were synthesized and their radical scavenging activities were evaluated and compared with that of the parent fisetin molecule. Among the methyl derivatives, 5′-methyl fisetin, in which the inherent planar structure of fisetin was retained, exhibited the strongest radical scavenging activity. Introduction of methyl substituents may be effective for the enhancement of various biological activities of antioxidants, particularly radical-scavenging activity. 相似文献
